Indication/Uses:
Amlodipine, branded as Amlosaph, is a calcium channel blocker that is commonly prescribed for the following conditions:
Hypertension (High Blood Pressure):
Amlodipine helps to lower blood pressure by relaxing the blood vessels, allowing blood to flow more easily. Lowering high blood pressure helps prevent strokes, heart attacks, and kidney problems.
Angina (Chest Pain):
It is used to prevent certain types of chest pain (angina). By relaxing the blood vessels and improving blood flow to the heart muscle, it helps to relieve chest pain and reduce the frequency and severity of angina attacks.
Coronary Artery Disease:
Amlodipine can be used to improve survival and reduce the risk of heart attack in patients with certain types of coronary artery disease.
Dosage
Typical Starting Dose:
The initial dose for adults is usually 5 mg once daily, which may be increased to a maximum dose of 10 mg once daily depending on the patient’s response and tolerance.
Mechanism of Action
Amlodipine belongs to the class of drugs known as calcium channel blockers. It works by inhibiting the movement of calcium ions into the smooth muscle cells of the heart and blood vessels. This results in the dilation of blood vessels and a reduction in blood pressure. It also decreases the heart’s workload by reducing the force of contraction and oxygen demand.
Administration
Oral Use:
Amlodipine tablets are taken orally, with or without food. It is important to take it at the same time each day to maintain an even level of the medication in your blood.
Side Effects
Common side effects may include:
Swelling (edema) of the ankles or feet
Headache
Dizziness
Flushing (feeling of warmth)

Precautions
Allergy:

Severe Aortic Stenosis:
Use with caution in patients with severe aortic stenosis as it can lead to increased risk of hypotension.
Hepatic Impairment:

Interactions
Amlodipine may interact with other medications, such as:
Other antihypertensive agents (increased risk of hypotension)
Simvastatin (may increase the risk of simvastatin side effects)
Grapefruit juice (can increase the blood levels of amlodipine and exacerbate side effects)

Periodic liver function tests may be advised for patients with liver disease.
